Abstract

Freeze-drying has usually been employed as preservation method for proteins. However, in spite of the low moisture coment, sorne degradarion reaccions as molecular aggregacion may take place during storage. If sugars are present, non enzymatic browning reactions have also been observed. Differential Scanning
Calorimerry (DSC) has been used for measuring glass transition temperature (Tg). Above this temperature, the product loses its rigidity and the vitrous condition turns sticky. Then, the degradation rate is ofte.n accelerated. The kinetics of degradation for most freeze-dried products stored at temperature above Tg is higher than expected when Arrhenius plot is employed. Then, it is suggested to use Williams-Landel-Ferry (WLF) equation. In this paper two
freeze-dried proteins (albumin and gammaglobulin alone or with glucose) are studied by means of DSC. The effect of storage temperature on molecular aggregation and non enzimatic browning are also srudied. The results are discussed in terms of WLF and Arrhenius k inetics.
