Abstract

The simultaneous detection in uríne of cocaine ( COC) and tbe main biotransformation products benzoylecgonine (BE), ecgonine methyl ester (EME) and ecgonine (EG) is a reliable way of confirmation of COC use. The purpose of this paper is the validation of qualitativc and quantitative method to simultaneous analysis in uríne of COC, cocaethylene (COCET) and tbe BE, EME and EG trimethylsilyl derivatives (BSTFA) with lidocaíne (LIDO) as internal standard method involves a solid-phase extraction (SPE) proce­dure using Evidex eartridges, and gas chromatographlc system, equipped witb DB-1 widebore column  (15 m x 0.53 mm x 3 µm),  operated with temperature pro from 60 C (heldfor2.rnin) to 140 Cat 20 C/min thentol70 C at 4 C/min, then to 208 ºC ( 40 ºc /min), then to 214ºC at 6°C/min (held for 6 min), then to 244ºC at 30ºC/min (he!d for 10 min), with spedfic nitrogen-phos­pborus detector (NPD), and N2 as earrier gas ( 4 mL/min). The calibrationgraph was linear from 406-4 060, 224-2 240, 108-1 088, 116-1167, 184-1840 ng/mL to EME, EG, COC, COCET and BE respectively. The relative standard deviation of response factors wa.s < 5, % in al! cases. Recoveries of spiked samples were 39, 62, 92, 85 and 92 % to EME, EG, COC, COCET and BE respectively. The
method was validated for use with urine samples. This is theJWst report of simultaneous SPE of COC and its main metabolites with Evidex1M cartridges.
