A MODEL OF CHARGE INTERACTION IN QUINOLONES

Abstract

Shen's model antibacterial action mechanism ofqui­nolones was analized . A QSAR srudy was developed wich che net charges at severa! positions of the quinolone moiety in a training set of 18 molecules. Four equations which correlate the net charges with the biological activity, measured against strains of E. coli H560 were obtained . The model was validated with a test set of 16 other structures, and was also employed to predict the more active conformations of a quiral compound. The relative relevance of ring-ring association on the binding with ADN and ADN-gyrase inhibition, is discussed.
