Nueva metodología para la producción de celulasas en Cuba

Abstract

The obtainment of enzymatic crudes able to carry out an outstanding activity on the substrate to convert it into a useful product is a
working line with great expectations. It is possible to use a protoplast production as a breeding way of used microorganisms. Crudes obtained from Aspergillus sp. strains through their cellulolitic activities in wild strains and in those submitted to the improvement with an alternative method were evaluated. Formic acid is included in the method as a possible hydrolytic agent. Some biochemical tests to characterize crudes were developed and individual reducer sugars were determined by two methods (C. and C1 activities) in crudes coming from wild individual strains and in their mixed crudes. Similar surveys were followed with crudes resulting from the growth of joinings of previousiy treated strains with formic acid, exposed or not to ultraviolet light, and with or without polyethyleneglycol additions. Proteincontents were also determined. Results of treatments with formic acid revealed a hydrolytic activity able to produce breaking conidia cell wall in both strains. The cellulolitic activity corroborated treatment advantages
with regard to individual cultures and mixed crudes because it was possible to release reducer sugars of 0,095 5; 0,211 1; 0,195 2 and 0,2218 mg/mL. These results also presuppose to have found an economic and efficient new methodology, for cellulases production in Cuba. Each crude includes an effective cellulolitic activity in the obtaining of plant protoplasts.
