Adding Pol-10 to the System Diagnostic Kit DIRAMIC and its influence on growth and study of susceptibility to antibiotics of urinary pathogens

Abstract

The addition of a linear polysaccharide polymer, named Pol-10, to the culture media of the DIRAMIC system's diagnostic kit was evaluated ;,this system is used in bacterial susceptibility test to antibiotics . The polymer Pol-10 was selected because it had no  undesirable effects on microorganisms and for its stability to bacterial enzymes . Three different polymer concentration ranges were assayed (0,05-0,15 ; 0,16-0,25 y 0,26-6,35 %) added to the M±eller-Hinton Broth . The incorporation of Pol-10 .at 0,05-0,15 % to the culture media, counteracts the inhibitory effect in the microbial growth of catabolic products accompanying the inoculli and therefore increases the indexes of microorganisms tested obtained ; furthermore, detection time of infections in clinical specimens may be shortened . The synergic effect between catabolites and antimicrobial activity was also eliminated, decreasing the false sensitive results from 15,2 to 3,9 % in 329 cases from clinics . Correspondence levels of DIRAMIC system methodology with the Bauer-Kirby
method varied from 78 .2 to 83 % .
